Body Clock and Sleep Problems in Inherited Polycystic Kidney Disease

Updated

Abstract

Circadian disruption in autosomal dominant polycystic kidney disease (ADPKD) may accelerate cyst growth and hypertension.

  • Genetic mutations in PKD1, PKD2, and other minor genes are established factors in ADPKD.
  • Circadian rhythms regulate important kidney functions such as glomerular filtration rate and sodium handling.
  • Disruption of circadian rhythms may activate sympathetic nervous system responses and dysregulate the renin-angiotensin-aldosterone system.
  • Increased levels of vasopressin and activation of the mTOR pathway are associated with accelerated disease progression.
  • Chronotherapy and sleep hygiene interventions could mitigate the effects of circadian misalignment in ADPKD.
