Circular RNA vaccines carrying fused Mpox virus proteins A35R-M1R and B6R-A29L trigger strong and lasting protective immunity in mice

Abstract

All bivalent mpox virus circRNA vaccines could provide 100% protection against lethal vaccinia virus challenge in mice.

  • The two constructed bivalent vaccines, cirBA and cirAM, encode different antigen combinations.
  • Both vaccines induced strong and lasting immune responses specific to mpox virus antigens.
  • When administered at low doses, the vaccines still conferred full protection against lethal infection.
  • At a higher dose, the vaccines maintained long-term immunity for at least 40 weeks.
  • The cirBA vaccine demonstrated superior protective effects compared to the other vaccine formulations.
