Abstract

Circular RNAs (circRNAs) may play critical roles in the pathogenesis of osteoarthritis (OA) by regulating gene expression.

  • CircRNAs are involved in multiple pathological processes associated with OA, including extracellular matrix degradation and chondrocyte apoptosis.
  • They may modulate signaling pathways such as NF-κB, PI3K/AKT, PTEN, and SIRT, which are important in OA development.
  • CircRNAs can act as either pathogenic amplifiers of cartilage degeneration or protective regulators that support chondrocyte survival.
  • They are associated with epigenetic regulation through DNA methylation and may facilitate intercellular communication within the joint via exosomes.
  • Advances in RNA engineering and delivery systems could enable clinical applications targeting circRNAs, though challenges in mechanistic understanding and delivery remain.
