Blood levels of TNF-α, CCL2, and IL-6 are linked to biological aging and specific DNA changes in aging mice

Updated

Abstract

Significant positive associations were found between epigenetic age and circulating TNF-α, CCL2, and IL-6 in older mice.

  • Epigenetic age was estimated using the multi-tissue DNAge panel, covering approximately 2000 DNA sites.
  • TNF-α was the only inflammatory mediator that maintained a significant independent association with epigenetic age in age-adjusted models.
  • Targeted analysis revealed that TNF-α was linked to a greater number of cytokine-related DNA sites compared to CCL2 and IL-6.
  • Stronger associations were detected in female mice regarding the relationship between TNF-α and epigenetic changes.
  • Exploratory analysis suggested positive correlations between TNF-α levels and two inflammation-related microRNAs, miR-155 and miR-21.
