Balancing binding and release in mRNA delivery using branched polyethyleneimine polymers of different sizes

Abstract

The optimal N/P ratio for full mRNA complexation was found to start at 5, regardless of the molecular weight of branched polyethylene-imine (PEI).

  • In SKOV-3 cells, 10 kDa PEI showed the highest transfection efficiency among the tested molecular weights.
  • 1.8 kDa bPEI/mRNA polyplexes did not successfully escape endosomal compartments.
  • 25 kDa bPEI/mRNA polyplexes were able to escape endosomes but demonstrated less mRNA dissociation in the cytosol.
  • In HeLa cells, both increasing the N/P ratio and the molecular weight of bPEI enhanced cellular uptake and transfection efficiency.
  • Transfection efficiency is likely limited by endosomal escape and the release of functional mRNA, which varies by cell type.
