Continuous Purification of Lab-Made mRNA Using Advanced Membrane Technology

Essence

High-performance countercurrent membrane purification may support continuous purification of in vitro-transcribed mRNA.

Evidence

A platform experiment with two model mRNA constructs removed 99.7% of CTPs at 60 min, ran for more than 24 h without fouling, and maintained >97% mRNA yield with >94% Proteinase K removal.

Caveat

Proteinase K removal fell when mRNA was present because of protein-mRNA complexation, and the study used model impurities rather than full manufacturing streams or clinical products.
