Cornuside may improve inflammation-related memory problems by boosting cell cleanup through Sirt1

Abstract

Cornuside improved cognitive performance in Alzheimer's disease mice and inhibited NLRP3 inflammasome activation.

  • Cornuside treatment (3, 10, and 30 mg/kg) for 2 weeks led to significant behavioral improvements in various cognitive tests.
  • The compound reduced levels of inflammatory markers associated with NLRP3 inflammasome activation in LPS-induced mice.
  • In vitro studies showed that cornuside similarly inhibited NLRP3 inflammasome activation in LPS-stimulated BV2 cells.
  • Cornuside increased the expression of Sirt1 and promoted autophagy, indicated by decreased SQSTM1/p62 and increased LC3BII.
  • Inhibition of autophagy with 3-MA and Sirt1 with EX527 negated the beneficial effects of cornuside on NLRP3 inflammasome activation.
