Attaching immune-activating molecules to lipids improves mRNA vaccine delivery and boosts immune cell activation

Updated

Abstract

A DSPE-CpG conjugate achieved 97.1% loading efficiency onto mRNA-LNPs.

  • The covalent conjugation platform integrates TLR9 agonists with phospholipids to improve immune response.
  • Enhanced cellular uptake was observed through clathrin-mediated endocytosis.
  • The system improved lymph node targeting of the adjuvant and antigen expression in vivo.
  • In the SARS-CoV-2 model, high levels of IFN-γ⁺CD8⁺ T cells and elevated specific IgG and neutralizing antibody titers were induced.
  • In the HBV model, there was a significant increase in antigen-specific CD107a and FasLCTLs.
  • Acute toxicity tests indicated no significant adverse effects.
