Fast testing of CRISPR guide RNAs using improved cell delivery in maize

Abstract

An optimized CRISPR/Cas9 system achieved 17.88 × 10⁶ viable maize protoplasts per gram fresh weight.

  • Protoplast isolation and transfection protocols were improved to enhance recovery and viability.
  • Transfection efficiency reached approximately 50% using 10 µg of plasmid DNA, without significant gains at higher DNA inputs.
  • Viability of protoplasts was maintained for up to seven days post-transfection, facilitating extended downstream applications.
  • Editing efficiencies for nine gRNAs targeting three floral repressor genes varied from 0.4% to 23.7% across four maize genotypes.
  • This system allows for rapid in vivo validation of gRNA activity, reducing assay time from months to days.
