Cycloastragenol may lengthen lifespan and boost stress resistance in worms by reducing insulin signaling and activating immune defense through p38 MAPK

Abstract

Cycloastragenol (CAG) at 0.2 μM extended mean lifespan by 30.64% in the Caenorhabditis elegans model.

  • CAG significantly improved aging-associated traits, including locomotion and stress resistance.
  • Treatment with CAG reduced lipofuscin accumulation and enhanced immunity against Pseudomonas aeruginosa.
  • CAG promotes the movement of the FOXO transcription factor DAF-16 into the nucleus.
  • Increased activities of antioxidant enzymes were observed with CAG treatment.
  • CAG may extend healthspan through a mechanism that inhibits the Insulin/IGF-1 Signaling pathway and activates the p38 MAPK axis.
