Dasatinib and Quercetin may improve bone loss in type 2 diabetes by changing blood chemicals and gut bacteria

Updated

Abstract

Essence

Dasatinib plus quercetin improved features in mice alongside shifts in gut microbiota and serum metabolism.

Evidence

This was a preclinical mouse study of type 2 diabetic osteoporosis that used micro-CT, histology, gut microbiome profiling, and serum metabolite analysis to show better bone-fat balance, more short-chain-fatty-acid-producing bacteria, fewer proinflammatory bacteria, higher amino acid and short-chain fatty acid metabolism, and lower cholesterol and triglycerides after treatment.

Caveat

Because the evidence comes from a mouse model with multi-omic associations, it does not prove which microbiome or metabolic changes drive the bone effects or whether the results will translate to humans.

What this is

  • This research investigates the effects of Dasatinib and Quercetin (D + Q) on () in a mouse model.
  • is characterized by reduced bone density and increased fracture risk due to metabolic dysregulation from type 2 diabetes.
  • The study examines how D + Q treatment influences gut microbiota composition and serum metabolite profiles, potentially alleviating bone loss.

Essence

  • D + Q treatment significantly improved bone health in mice by enhancing gut microbiota and serum metabolites, while reducing weight gain and hyperglycemia.

Key takeaways

  • D + Q treatment reduced fasting blood glucose (FBG) from ~27.43 mmol/L to ~21.29 mmol/L, indicating improved glycemic control.
  • Bone mass was restored in mice after 6 weeks of D + Q treatment, with significant increases in trabecular bone thickness and reductions in bone loss.
  • D + Q treatment modulated gut microbiota, increasing beneficial bacteria and decreasing proinflammatory species, which may contribute to improved bone metabolism.

Caveats

  • The study's findings on metabolomics are exploratory and require further validation to establish causal relationships.
  • Results were derived from male mice, limiting generalizability to female physiology and necessitating future studies to include both sexes.

Definitions

  • Type 2 diabetic osteoporosis (T2DOP): A metabolic bone disorder linked to type 2 diabetes, characterized by reduced bone density and increased fracture risk.
  • Senolytics: Compounds that selectively induce death of senescent cells, potentially improving age-related conditions.
