Lower Energy Use and Protein Helpers Support Aging Fibroblast Survival, with Pyruvate Dehydrogenase as a Strong Target to Remove Them

Updated

Abstract

Essence

Senescent fibroblasts appeared especially dependent on TCA-cycle input and chaperone activity for survival.

Evidence

This proteomic and experimental preclinical study profiled ionizing-radiation-induced senescent human BJ fibroblasts and tested inhibition of pyruvate dehydrogenase, GLS1, and Hsp90 in senescent cells and aged mice.

Caveat

The senolytic findings come from cell models, therapy-induced senescent cancer cells, and aged mice, not human clinical testing.
