Desflurane raises key protein levels and lowers clock protein in memory areas, while Sorafenib reduces thinking problems in old mice

Updated

Abstract

Desflurane exposure in aged mice decreased CLOCK protein levels and was associated with cognitive deficits.

  • Exposure to 2% desflurane for 2 hours increased total c-Raf, ERK1/2, and MSK1 protein levels in the hippocampus.
  • Decreased CLOCK protein levels in the aged hippocampus correlated with increased cellular senescence markers and cognitive impairments.
  • RAF manipulation in hippocampal neurons bidirectionally altered CLOCK protein levels, with knockdown restoring CLOCK levels.
  • In aged mice, pretreatment with the RAF inhibitor sorafenib improved cognitive performance and reduced markers of cellular senescence.
