Different responses of two prostaglandin receptors in immune brain cells activated by bacterial molecules

Abstract

PGE production after LPS was found to be COX-2-dependent.

  • PGE reduces the production of the inflammatory protein TNF-α in glial cells after LPS exposure.
  • Microglia express higher levels of EP4 and EP2 receptors compared to astroglia.
  • Activation of EP4 or EP2 receptors with specific drugs decreases LPS-induced TNF-α in microglia.
  • Antagonizing EP4 prevents the anti-inflammatory effect of PGE in naïve microglia, indicating it as the main target.
  • The expression of EP receptors changes during microglial activation, with EP4 levels decreasing and EP2 levels increasing 24 hours after LPS treatment.
