Diosmin reduces colitis by stopping a mixed cell death in gut lining cells and balancing gut bacteria and their chemicals

Abstract

Diosmin (DIO) significantly improved colitis symptoms in mice, as shown by reduced weight loss, lower Disease Activity Index scores, and preserved colon length.

  • DIO treatment diminished inflammation and tissue damage in the colon of DSS-induced colitis mice.
  • Mechanistically, DIO suppressed genes and proteins associated with a type of cell death known as PANoptosis.
  • DIO maintained the integrity of the intestinal barrier in human colonic epithelial cells exposed to inflammatory conditions.
  • The treatment altered gut microbiota composition, increasing beneficial bacteria such as Ruminococcus and decreasing harmful Proteobacteria.
  • Metabolomic analysis indicated changes in important metabolic pathways, including those related to flavonoid and steroid hormone production, which were linked to changes in gut microbiota.
