Different molecular ways glioblastoma cells develop resistance to temozolomide

Abstract

TMZ resistance may involve different molecular mechanisms in glioma cells.

  • In TMZ-resistant LN-18 cells, there was a strong increase in levels of the DNA repair protein MGMT.
  • TMZ resistance in MGMT-negative LNT-229 and LN-308 cells was not linked to new MGMT expression.
  • Resistant LNT-229 cells showed a decrease in several DNA mismatch-repair proteins.
  • Silencing specific DNA mismatch repair proteins in parental LNT-229 cells led to a TMZ-resistant phenotype.
  • No clear resistance mechanism was found in LN-308, aside from decreased methylation of LINE-1 repetitive elements.
