Abstract

35% and 40% reductions in elevated serum ALT and AST levels were observed in MASH mice following disulfiram administration.

  • Disulfiram may reduce lipid accumulation and lipotoxicity in liver cells exposed to free fatty acids.
  • In a mouse model of metabolic dysfunction-associated steatohepatitis, disulfiram significantly reduced hepatic triglyceride content from 1 to 0.5 µg/mg protein.
  • Disulfiram administration markedly decreased hepatic steatosis, inflammation, fibrosis, and oxidative stress.
  • RNA sequencing identified Aurora kinase A as a central gene modulated by disulfiram, linked to changes in autophagy-related markers.
  • Inhibition of Aurora kinase A by disulfiram is associated with enhanced autophagy and reduced liver damage.
