DNA methylation linked to aging of lung repair cells in COPD

Abstract

A total of 19 eQTMs were identified for COPD-associated senescence genes.

  • Cellular senescence is linked to chronic obstructive pulmonary disease (COPD), with increased levels observed in lung fibroblasts from patients.
  • Gene expression analysis revealed higher levels of CDKN1A, CDKN2A, and CDKN2B, while LMNB1 expression was lower in COPD-derived fibroblasts compared to controls.
  • Seven specific DNA regions (CpG sites) showed significant differences in methylation levels between COPD patients and matched controls.
  • Treatment with 5-Aza-2'-dC resulted in global demethylation of fibroblasts and increased senescence, supporting the connection between altered DNA methylation and senescence.
  • The identified CpG sites may serve as potential epigenetic regulators for the senescence genes CDKN1A and LMNB1 in COPD.
