How the structure of a designed mRNA vaccine affects immune response against key proteins of tuberculosis bacteria

Pattern 3 (PPE68-IrtA-PE9) achieved the highest antigenicity score of 0.6122 among the evaluated vaccine constructs.

  • The multi-epitope vaccine constructs were designed using an immunoinformatics-driven strategy.
  • Pattern 3 demonstrated a high abundance of B-cell epitopes, scoring 0.865.
  • Predicted binding affinities to the TLR4/MD-2 complex and TLR2 were ΔG = -12.2 kcal/mol and ΔG = -10.4 kcal/mol, respectively.
  • In silico immune simulations indicated strong predicted T-cell responses and elevated levels of IFN-γ.
  • The codon-optimized mRNA exhibited a stable secondary structure with a ΔG of -2,217.20 kcal/mol.
  • The results suggest that the arrangement of antigen domains may influence immunogenicity and structural stability.
