Creating a human stem cell line with controllable p16INK4a gene activation using doxycycline

Updated

Abstract

Doxycycline treatment induces dose-dependent increases in p16INK4a expression in a newly established human embryonic stem cell line.

  • A doxycycline-inducible p16INK4a knock-in human embryonic stem cell line was created by targeting a specific safe-harbor locus.
  • The cell line maintained essential characteristics, including pluripotency marker expression and tri-lineage differentiation potential.
  • Normal karyotype was confirmed in the generated cell line.
  • This model may assist in studying p16INK4a-associated cellular senescence across various stem cell-derived lineages.
