Mitochondrial Splitting by Drp1 Helps Control Mineral Transport and Mitochondrial Balance During Bone Formation

Updated

Abstract

Drp1 inhibition led to a 59.2% reduction in ATP production in human mesenchymal stem cells during osteogenic differentiation.

  • Phosphorylation of Drp1 at Ser616 increased while phosphorylation at Ser637 decreased after osteogenic induction.
  • Increased mitochondrial fission produced fragments approximately 450 nm in diameter, aiding in the transport of calcium phosphate precursors.
  • Knockdown of Drp1 caused abnormal elongation of mitochondria, accumulation of calcium phosphate precursors, and reduced ATP levels.
  • Mineralization area decreased to 36.7% following Drp1 inhibition, alongside suppressed expression of osteogenic genes RUNX2 and ALPL.
  • In a zebrafish model, Drp1 inhibition similarly reduced craniofacial mineralization and downregulated osteogenic gene expression.
