Drug Seeking and Avoidance Strengthen Connections on Different Groups of Reward System Neurons

Abstract

Mice that refrained from cocaine seeking in an extinguished context showed a selective increase in AMPA/NMDA ratios in D2-expressing neurons.

  • Transient potentiation of glutamatergic synapses in the nucleus accumbens core is linked to drug-seeking behavior.
  • D2-expressing neurons exhibited elevated AMPA/NMDA ratios when mice refrained from seeking cocaine after extinction training.
  • In the absence of extinction training, the presence of drug-associated cues led to increased AMPA/NMDA ratios in D1-expressing neurons.
  • Both D1 and D2 neurons showed potentiation in response to cue-induced cocaine seeking following extinction training.
  • These findings suggest distinct roles for D1 and D2 neuron populations in the motivation to seek or refrain from drugs.
