Liver cancer vaccine using two immune boosters and fat-based carriers to boost treatment

Abstract

L1-LNPs/HGPC3C mRNA achieved 80.4% tumor suppression in prophylactic models.

  • Glypican-3 (GPC3) is a potential target for immunotherapy in liver cancer due to its tumor-specific expression.
  • A dual-adjuvanted mRNA vaccine using L1-based lipid nanoparticles was developed to enhance immune responses.
  • The engineered L1-LNPs demonstrated 4-fold greater local expression compared to another lipid nanoparticle formulation.
  • Mechanistic analyses indicated that HSP70 improved cellular immune responses, while C3d enhanced humoral immune responses.
  • In therapeutic models, the L1-LNPs/HGPC3C formulation significantly outperformed non-adjuvanted vaccines.
  • Combining this vaccine with PD-L1 blockade further increased tumor suppression and immune cell infiltration.
