The optimized mRNA vaccine construct RH5-T2A-CSP (Mal05) conferred 100% sterile protection in murine models against Plasmodium berghei.
- Mal05 targets both the pre-erythrocytic and blood stages of Plasmodium falciparum.
- High-titer, durable antibodies were elicited that can inhibit sporozoite invasion and erythrocytic growth in vitro.
- The vaccine induced strong Th1-biased CD4⁺ and CD8⁺ T-cell responses.
- Dual-antigen mRNA-LPP vaccination may establish a potent immunological barrier by integrating neutralizing antibody activity with cellular immunity.
