A dual compartment lipid carrier system may enhance antitumour efficacy by restoring cellular energy levels and modifying metabolic processes.

  • Lactate driven immunosuppression is linked to reduced levels of intracellular nicotinamide adenine dinucleotide.
  • Restoring nicotinamide adenine dinucleotide through lipid nanoparticles may improve T cell function.
  • Targeting the endoplasmic reticulum allows for specific export of dichloroacetate, which could help deplete lactate in a localized manner.
  • This approach may extend the persistence and reactivation of natural killer cells in the tumour environment.
  • The strategy of exocytosis co-option could potentially enhance the effectiveness of various adoptive cell therapies.
