Vaccine using two glycolipids allows choice of delivery method to target lung tumor spread

Updated

Abstract

Inhaled SG-LNP vaccines achieved 100% protective efficacy against pulmonary metastasis in a melanoma model.

  • The PEG-free mRNA platform, co-modified with sialic acid-dimyristoyl-glycerol and monosialotetrahexosylganglioside, enhances targeting of antigen-presenting cells.
  • Inhaled SG-LNP vaccines lead to significant local immune activation, reducing melanoma infiltration in the lungs compared to conventional PEG-modified LNPs.
  • Systemic administration of SG-LNP vaccines results in greater expansion of circulating antigen-specific T cells and natural killer cells.
  • This systemic mobilization may intercept circulating tumor cells before they can colonize the lungs.
  • The findings suggest that coordinated systemic immune responses could be more effective than localized activation for preventing organ-specific metastasis.
