Lipid nanoparticles targeting two sites to deliver TET3 siRNA and change the tumor immune environment in liver cancer

Updated

Abstract

TET3 siRNA@αmp-Lipo significantly suppressed tumor growth in hepatocellular carcinoma models.

  • Single-cell RNA sequencing revealed that TET3 is highly expressed in M2-polarized tumor-associated macrophages in HCC tissues.
  • TET3 enhances the activity of a specific transcription factor, which leads to CD8+ T cell exhaustion through a particular signaling pathway.
  • Dual-targeted lipid nanoparticles were engineered to deliver TET3 siRNA specifically to M2 macrophages.
  • In vitro results showed these nanoparticles effectively silenced TET3, inhibited M2 polarization, and restored CD8 T cell function.
  • In vivo studies indicated that the treatment reduced M2 macrophage infiltration and reversed CD8 T cell exhaustion.
