Abstract

Duhuo Jisheng Decoction (DHJS) significantly alleviated intervertebral disc degeneration (IVDD) by inhibiting nucleus pulposus cells (NPCs) pyroptosis.

  • Acacetin (ACA), a key component of DHJS, directly binds to MAPK1 and inhibits its expression.
  • The inhibition of MAPK1 by ACA relieves its negative regulation on heme oxygenase 1 (HMOX1).
  • This interaction promotes mitophagy in NPCs and suppresses pyroptosis.
  • In vivo validation showed that ACA reversed collagen fiber disorder and proteoglycan reduction in IVDD rat models.
  • This study identifies the Acacetin-MAPK1/HMOX1 axis as a regulatory pathway linking mitophagy and pyroptosis in IVDD.
