How bone sensing of movement slows aging and may extend lifespan

Updated

Abstract

Reduced mechano-sensing in osteocytes accelerated whole-body aging in mice.

  • Dynamic tibial axial loading (DTAL) extended the healthspan and lifespan of mice by 9.4% or 79.5 days.
  • DTAL reversed bone marrow immunosenescence and promoted factors associated with anti-aging.
  • Pro-aging and pro-inflammatory factors were downregulated following DTAL treatment.
  • Pigment epithelium-derived factor (PEDF) was identified as a novel mechano-responsive anti-aging factor linked to healthy aging in humans.
  • Bone mechano-sensing plays a crucial role in the aging process of the whole body.
