Synthetic miRNA carriers targeted to blood vessel and iron transport proteins with potential stroke therapy

Abstract

Small sized, uniform lipid nanoparticles (LNPs) were developed to deliver miRNAs targeting post-stroke inflammation.

  • The LNPs demonstrated high miRNA encapsulation efficiency.
  • Functionalization with specific antibodies allowed targeting of inflamed cerebral blood vessels.
  • In vitro assays showed that the LNPs reduced interleukin-6 production in human brain endothelial cells.
  • In a mouse model, low doses of targeted LNPs reduced infarct volume and suggested improvements in neurological deficits.
  • High doses of targeted LNPs did not produce the same beneficial effects.
