Easily Creating Gene-Edited Onions Using New Techniques

Abstract

A total of 47 onion plants with edited AcDMR6 alleles were produced using a transient gene expression method.

  • The technique allowed for the recovery of gene edited onion lines without relying on stable transformation.
  • Co-delivery of plasmids for antibiotic resistance and plant developmental regulation alongside Cas9/sgRNA RNPs enhanced gene editing.
  • Up to 12% of regenerated plants exhibited edited alleles of AcDMR6, a significant improvement compared to zero editing in controls.
  • The method may be applicable to other crop species, suggesting broader potential for crop gene editing.
