Ecliptasaponin A reduces colitis in mice by blocking harmful immune cell changes through the JAK2/STAT3 pathway

Abstract

Ecliptasaponin A (ESA) treatment significantly reduced disease activity index in mice with DSS-induced inflammatory bowel disease.

  • ESA treatment alleviated body weight loss and colon shortening in mouse models.
  • It significantly decreased the expressions of inflammatory mediators TNF-α, IL-6, and iNOS in the colon.
  • ESA protected intestinal barrier integrity by upregulating ZO-1 and claudin-1 expressions.
  • Treatment decreased M1 macrophage polarization indicated by reduced CD86 expression in mesenteric lymph nodes.
  • ESA also lowered levels of phosphorylated JAK2 and STAT3 proteins in both mouse models and cultured macrophages.
