Editing Probiotic Bacteria to Reduce Hemolysin Production

Abstract

A 50 bp deletion and the introduction of a stop codon in the hlyIII gene were achieved in L. plantarum and L. brevis using a Cas9-gRNA ribonucleoprotein complex.

  • Genome editing was successfully performed on the hlyIII gene in Lactobacillus strains.
  • The ΔhlyIII mutants demonstrated reduced hemolytic activity, with decreases in free hemoglobin levels of 27.0%, 74.3%, and 5.0% for L. plantarum WCFS1, L. plantarum SPC 72 - 1, and L. brevis SPC-SNU 70 - 2, respectively.
  • Strain-dependent differences in hemolytic activity suggest varying roles of the hlyIII gene in different Lactobacillus species.
  • The editing system developed may facilitate broader applications in probiotic engineering by reducing reliance on plasmids and antibiotics.
  • Further validation is necessary to fully understand the functional implications of hlyIII in hemolytic activity.
