How Duhuo Jisheng Decoction May Help Treat Spinal Disc Degeneration: Combining Network Analysis and Lab Experiments

Abstract

Duhuo Jisheng decoction (DHJSD) improved nucleus pulposus (NP) cell viability by 20% at a concentration of 20% after 24 hours.

  • DHJSD reduced the expression of inflammatory markers NLRP3, ASC, caspase-1, and IL-1β in NP cells compared to the pressure group.
  • Expression of protective factors collagen II, aggrecan, Sox-9, PINK1, Parkin, and LC3 was significantly increased in the DHJSD group.
  • The addition of a mitophagy inhibitor diminished the protective effects of DHJSD on NP cells.
  • Network pharmacology suggested that HIF-1α may be a key target for DHJSD in treating intervertebral disc degeneration (IDD).
  • Imaging and histopathological results indicated that DHJSD delayed the progression of IDD.
