Combined Elamipretide and NMN Treatment May Reduce Brain Damage After Stroke by Targeting TREM2 in Mice

Co-administration of SS-31 and NMN significantly reduced brain damage and neurological deficits in a mouse model of ischemic stroke (P < 0.0001).

  • The combination therapy of SS-31 and NMN demonstrated effects that were markedly superior to either treatment alone.
  • Transcriptomic profiling indicated that the therapy specifically altered innate immune and cell death pathways, resulting in significant downregulation of TREM2.
  • Inhibition of the NF-kB pathway was associated with decreased activation of TREM2, leading to reduced microglial activation and lower levels of pro-inflammatory cytokines.
  • Suppression of TREM2 expression helped rebalance the apoptotic signals in the brain by affecting the Bcl-2/Bax ratio.
  • Overexpression of TREM2 negated the neuroprotective effects of the combination therapy, suggesting its critical role in the observed outcomes.
