Improving Cell Delivery for Aptamer-Driven Ubiquitin-Based Breakdown of c-Met Protein

Updated

Abstract

The small molecule chloroquine enhances endosomal escape of the c-Met targeting aptamer SL1 more effectively than the peptide GALA.

  • Systematic mapping revealed that the c-Met targeting aptamer SL1 is predominantly retained in endosomes with minimal release into the cytosol.
  • Two agents were evaluated for their ability to facilitate endosomal escape, with chloroquine showing superior effectiveness compared to GALA.
  • An aptamer-based PROTAC was developed to promote c-Met interaction with an E3 ligase, leading to its ubiquitination.
  • Enhanced endosomal escape resulted in ubiquitin-mediated degradation of c-Met, inhibiting proliferation and migration of target cells.
  • This approach successfully suppressed tumor growth in a xenograft model.
