How cell-penetrating peptides escape cell compartments to deliver substances inside cells

Abstract

Endosomal escape of cargo delivered by cell-penetrating peptides (CPPs) was found to be very inefficient in a macrophage cell line.

  • The CPPs TAT, (RW), and CPP9 improved the cytosolic delivery of DINNN-containing peptides.
  • Enhancements in delivery were primarily due to increased uptake into endosomes, not improved endosomal escape.
  • Inclusion of the fluorophore sCy5 in peptide conjugates generally increased cytosolic uptake, although effects varied by CPP.
  • Confocal imaging supported findings from the endosomal escape quantitation assay, highlighting potential issues with dye effects in uptake studies.
  • The results suggest that CPPs facilitate cytosolic delivery mainly through membrane interaction and endosomal incorporation.
