RNA chemical changes in COVID-19 linked to how the virus affects the body

Abstract

In whole blood samples from COVID-19 patients, 1,201 pseudouridine (psU) sites were detected, many uniquely enriched compared to healthy controls.

  • Three key RNA modifications (5-methylcytosine, N6-methyladenosine, and pseudouridine) were profiled in the context of SARS-CoV-2 infection.
  • Condition-specific modification sites were identified, with distinct positional distributions primarily in 3' UTRs and final exons.
  • Functional annotation linked these RNA modifications to immune response, viral defense, inflammation, and stress.
  • Pathway analysis highlighted convergence on processes crucial for viral entry and immune activation, such as endocytosis and phagocytosis.
  • The findings suggest that RNA modifications may serve as biomarkers and therapeutic targets in COVID-19.
