Exendin-4 influences key cell signals in inflamed human ligament stem cells to encourage bone formation

Abstract

Exendin-4 (Ex-4) promotes osteogenic differentiation of periodontal ligament stem cells (PDLSCs) in an inflammatory environment.

  • LPS treatment increased the proliferation of PDLSCs but inhibited their ability to differentiate into bone.
  • Ex-4 reversed the effects of LPS by decreasing PDLSC proliferation and enhancing osteogenic differentiation.
  • Ex-4 elevated levels of key osteogenic markers, including Runx2, ALP, and Osx, while reducing pro-inflammatory cytokines TNF-α and IL-6.
  • The treatment with Ex-4 inhibited the nuclear translocation of NF-κB/p65, which is involved in inflammatory responses.
  • Co-treatment with Ex-4 and LPS resulted in decreased expression of β-catenin in the nucleus, suggesting modulation of Wnt signaling.
