Autophagy

Exercise causes a chemical change in WDR41 that starts cell cleaning and helps improve fatty liver disease

Updated

Abstract

Negative correlations were found between the expression and lactylation of WDR41 and human hepatic metabolic diseases.

  • WDR41 is identified as a pro-autophagy protein that undergoes lactylation during conditions that induce autophagy, such as exercise.
  • Lactylation of WDR41 at lysine 319 (K319) is facilitated by lactate dehydrogenase A (LDHA) when autophagy is activated.
  • Delactylation of WDR41 is mediated by sirtuin 2 (SIRT2).
  • Lactylated WDR41 enhances autophagy initiation by interacting with autophagy proteins ATG13 and RB1CC1/FIP200, promoting the assembly of the ULK1 complex.
  • WDR41 and its lactylated form at K319 are crucial for exercise-induced improvements in insulin sensitivity and lipid metabolism in the liver.
