Longer gaps between doses improve lasting protection of a self-boosting mRNA vaccine against pandemic H5N1 flu

Updated

Abstract

A Phase 1 trial involving 212 healthy adults demonstrated that the self-amplifying mRNA vaccine ARCT-2304 was well-tolerated and elicited strong immune responses.

  • Two doses of ARCT-2304 led to robust antibody responses against H5N1 hemagglutinin and neuraminidase.
  • An 8-week dosing schedule resulted in higher and more durable immune responses compared to a 4-week schedule.
  • Neutralizing and anti-NA antibodies remained significantly elevated 6-7 months after the second dose.
  • The vaccine exhibited mostly mild-to-moderate, transient adverse effects that were dose-dependent.
  • No safety concerns were identified during the trial.
