How a specific protein helps clear damaged cell parts to reduce liver cell death caused by dithiothreitol

Abstract

FAM134B downregulation accelerated hepatocyte apoptosis under ER stress conditions.

  • ER stress-related and autophagy-related proteins were elevated in BRL-3A cells after both short and long-term DTT treatment.
  • Co-immunoprecipitation demonstrated an interaction among FAM134B, calnexin, and LC3 in BRL-3A cells.
  • Immunofluorescence assays showed a decrease in autolysosomes following short-term DTT treatment, with an increase after long-term treatment.
  • Mitochondrial calcium levels and apoptotic rates significantly increased after short-term DTT treatment but decreased 48 hours later.
  • FAM134B-mediated ER-phagy is associated with the attenuation of the mitochondrial apoptotic pathway in hepatocytes.
