Abstract

FOCUS LNPs achieve ∼12-fold greater mRNA expression in fibrotic lung tissues compared to traditional delivery methods.

  • FOCUS combines ionizable lipids and fibroblast targeting ligands to enhance mRNA delivery.
  • These LNPs show ∼3-fold and ∼12-fold increases in mRNA expression in fibrotic liver and lung, respectively.
  • Fibroblast distribution is increased by ∼2-5-fold with FOCUS LNPs compared to organ- or ligand-only targeting.
  • Pentraxin-2 mRNA-loaded FOCUS LNPs reduce collagen deposition by 60-80% in murine models of fibrosis.
  • The treatment avoids systemic side effects, such as impaired wound healing, while suppressing fibroblast activation.
