How FCCP Triggers Mitochondrial Cleanup in Human Airway Muscle Cells

Abstract

Exposure of human airway smooth muscle cells to 1 µM FCCP for 6 hours induced mitochondrial depolarization and decreased intact mitochondria.

  • Mitochondrial depolarization led to the accumulation of PINK1 in the mitochondria.
  • PINK1 accumulation resulted in the phosphorylation of Parkin at serine 65.
  • Phosphorylated Parkin (pParkin<S65</S65>) initiated the ubiquitination of outer mitochondrial membrane proteins.
  • Increased colocalization of mitochondria with lysosomes was observed, indicating enhanced mitophagy.
  • Mitophagic flux was confirmed using a pH-sensitive mitochondrial reporter.
