Ferrostatin-1 may reduce spinal cord damage after blood flow loss and return through a protective cell pathway

Abstract

Intrathecal injection of ferrostatin-1 (Fer-1) improved neurological function and reduced BSCB leakage after spinal cord ischemia/reperfusion injury.

  • Ferroptosis is linked to neuronal death caused by spinal cord ischemia/reperfusion injury (SCIRI).
  • Fer-1 increased motor neuron counts and neurological function scores while decreasing neuroinflammation levels in the anterior horn.
  • SCIRI led to elevated levels of several proteins associated with ferroptosis, while Fer-1 ameliorated these changes.
  • Fer-1 enhanced the expression of p-ERK1/2 and SP1, which are involved in the ERK1/2/SP1/GPX4 signaling pathway.
  • Inhibitors targeting the ERK pathway reduced the increase in SP1 and GPX4 levels induced by Fer-1.
