Fibroblast aging and related secretions linked to outcomes in pancreatic cancer's tumor environment

Abstract

In a study of 90 patients with pancreatic ductal adenocarcinoma, high levels of IL-6-positive fibroblasts were associated with reduced postoperative survival.

  • Senescent fibroblasts in the PDAC stroma express the senescence marker p16 and the SASP factor IL-6.
  • A positive correlation exists between the expression levels of p16 and IL-6 in fibroblasts.
  • Increased IL-6 expression in fibroblasts is linked to poorer outcomes in patients.
  • High IL-6 expression and lymph node metastasis were identified as independent prognostic indicators.
  • Senescent fibroblasts enhanced the invasion, migration, and proliferation of pancreatic cancer cells in co-culture experiments.
