Fibulin-3 Controls a Protein That Prevents Aging in the Core Cells of Spinal Discs

Abstract

A negative correlation exists between intervertebral disc degeneration grade and fibulin-3 expression (r = -0.79, p < 0.001).

  • Higher degeneration grades of intervertebral discs are associated with increased senescence of nucleus pulposus cells (NPCs) (r = -0.87, p < 0.001).
  • As NPCs undergo more passages in culture, fibulin-3 expression decreases, while inflammatory cytokines and matrix metalloproteinases increase.
  • Adding exogenous fibulin-3 significantly inhibits NPC senescence and reduces inflammatory cytokine and matrix metalloproteinase levels.
  • Gene overexpression of fibulin-3 reduces NPC apoptosis and enhances cell proliferation, suggesting a protective role against senescence.
  • Fibulin-3 is linked to TIMP-3 regulation, which may inhibit matrix metalloproteinase activity and promote extracellular matrix synthesis.
