Protecting cartilage in osteoarthritis through cell cleanup controlled by FoxO1, PINK1, and Parkin triggered by Guzhi Zengsheng Zhitong decoction

Abstract

Guzhi Zengsheng Zhitong Decoction (GZZD) significantly improved knee joint health in a rat model of osteoarthritis.

  • GZZD reduced cartilage degradation and subchondral bone destruction in knee osteoarthritis.
  • Treatment led to lower scores on the Osteoarthritis Research Society International (OARSI) scale.
  • Serum levels of pro-inflammatory cytokines such as IL-1β, IL-6, and TNF-α were significantly decreased.
  • GZZD enhanced protective autophagy in chondrocytes and inhibited apoptosis and mitochondrial dysfunction.
  • The FoxO1/PINK1/Parkin pathway was activated by GZZD, restoring mitochondrial function in chondrocytes.
  • The study suggests a potential role for GZZD in developing therapies targeting FoxO1 for osteoarthritis.
