Basic Sex Differences in Cocaine-Related Changes in Two Types of Reward Neurons in the Mouse Brain

Abstract

Cocaine treatment altered neuronal excitability in female mice, shifting effects from D1R-expressing neurons to D2R-expressing neurons.

  • In males, cocaine reduced the excitability of D1R-expressing medium spiny neurons (D1R-MSNs) without affecting D2R-expressing medium spiny neurons (D2R-MSNs).
  • In females, D1R-MSN excitability changed across the estrous cycle, mirroring the effects observed in males following cocaine exposure.
  • Cocaine increased the frequency and amplitude of miniature excitatory postsynaptic currents (mEPSCs) in D2R-MSNs of females during estrus and amplitude in females during diestrus.
  • Males exhibited increased mEPSC amplitudes in both D1R- and D2R-MSNs after cocaine treatment, with no change in mEPSC frequencies.
