Galectin-3 may protect against prion disease by helping brain immune cells clear harmful proteins

Updated

Abstract

Galectin-3 (Gal3) expression was undetectable in healthy brains but became upregulated during prion disease progression.

  • Gal3 localized predominantly to a specific subset of reactive microglia in the thalamus as the disease advanced.
  • Increased Gal3 expression inversely correlated with homeostatic microglial markers, indicating a change to a reactive microglial phenotype.
  • Constitutive deletion of Gal3 accelerated clinical disease progression without affecting total prion accumulation or overall microglial activation.
  • Gal3 deficiency reduced microglial uptake of prions, leading to a lower intracellular-to-extracellular prion ratio.
  • Gal3 did not impair lysosomal activity or membrane integrity, suggesting its role is specific to phagocytosis rather than lysosomal function.
